About Jennie Sjöhamn

Research
My research is focused on membrane proteins and the characterization of their structure, function and interaction partners. I've used human water channels, aquaporins, to understand which factors that affect the overproduction of membrane proteins in the P. pastoris protein production system. The human aquaporin with the highest production yields, hAQP10, has been studied further as it showed a population heterogeneity which was found to be caused by a partial glycosylation. This glycosylation stabilizes the protein and site-directed mutagenesis that abolishes the glycosylation site give a homogenous sample but decreases the thermostability of the protein.

I've also worked on the protein purification optimization and construct design of a bacterial enzyme, ubiquinol oxidase, which is part of the E. coli energy generation in the electron transport chain.

By using X-ray crystallography we can study the structure of these proteins. We can also look at dynamics over small time scales within the protein with time resolved WAXS (Wide Angle X-ray Scattering).
